2024.06.05 11:16 cs3001 Important role of copper & vit D in schizophrenia (transporters) (Vitamin D and A / thyroid)

Important role of copper & vit D in schizophrenia (transporters) (Vitamin D and A / thyroid)
Copper looks to be highly significant in schizophrenia,
where blood levels can be higher than normal BUT brain levels lower in copper regardless (due to defective copper transport)
so problem can be with transport typically instead of levels, leading to functional deficiency in some brain areas at least:
ATP7A and B, and CTR1 are involved in copper transport
insightful studies
https://www.ncbi.nlm.nih.gov/pmc/articles/PMC6424639/
https://www.ncbi.nlm.nih.gov/pmc/articles/PMC7987889/
These results provide the first evidence of disrupted copper transport in schizophrenia SN that appears to result in a copper-deficient state. Furthermore, copper homeostasis may be modulated by specific dysbindin isoforms and antipsychotic treatment.
Interestingly, decreasing copper activity through by inhibiting transporters or administering the copper chelator cuprizone to mice produces demyelination, altered neurotransmitters, and decreased oligodendrocytic protein expression (Gokhale et al. 2015; Gregg et al. 2009; Herring and Konradi 2011). Additionally, **reduced copper activity results in schizophrenia-like behavioural impairments**, such as deficits in novel object recognition, spatial memory tasks, pre-pulse inhibition, social interaction, and anxiety
Additionally, **schizophrenia patients exhibit decreased metallothionein and glutathione, responsible for intracellular copper chaperoning and transport, that are rescued with antipsychotic treatment** (Xuan et al. 2015; Do et al. 2000)(Figure 1B)
In addition to those mentioned previously, **lack of copper also exacerbates NMDA-mediated excitotoxic cell death in primary hippocampal neurons**
Given the **excess of copper in the blood of schizophrenia patients** and the increasingly mounting evidence of an abnormal or “leaky” BBB in psychosis, we **hypothesize that schizophrenia patients exhibit excess blood copper due to faulty copper transport** across the blood brain barrier (BBB). **This abnormality would result in a copper-deficient brain state** and contribute to the cognitive and behavioral deficits observed in schizophrenia. Our results suggest extracellular copper binding and transport into the cell **via CTR1 is impaired in schizophrenia**, and not rescued with treatment
when subdivided by medication status, the N-terminus **[ATP7a] protein levels were significantly lower** in unmedicated patients versus medicated patients (p=0.02) and controls
Analysis of treatment status revealed that C-terminal ATP7A protein levels were significantly increased only in medicated patients versus controls
![29a20092-8340-4477-8b28-76644773a3ca-image.png](/assets/uploads/files/1717572133634-29a20092-8340-4477-8b28-76644773a3ca-image.png)
https://preview.redd.it/7eedo54owp4d1.png?width=1155&format=png&auto=webp&s=bdc22f906a421ecb1dd112b136a112d2ada08fe6
Copper transport proteins ^ ,
**people with schizophrenia not on medication have much lower n-terminus ATP7A expression. much lower transmembrane CTR1 expression. lower ATP7b expression.**
https://preview.redd.it/h4n00lhpwp4d1.png?width=833&format=png&auto=webp&s=2a39027c680356921597795e26ae91f7bfc78640
**and less copper in the substantia nigra brain area tested for copper content**
**[So this suggests another likely helpful area of focus is finding stuff that increases ATP7a and especially CTR1 expression]**
**3 basic things,**
Vitamin A or thyroid hormone could help increase atp7a and vitamin D could help increase ctr1 (but need to be balanced)**
**Interestingly there's been some mention of how schizophrenia has some commonalities with vitamin a excess or deficiency**
(i) neurological congenital abnormalities reported in some schizophrenia cases are **comparable to those observed with vitamin A deficiency** or altered at-RA
(either deficiency or maybe excess also, as maybe overload could take away from vitamin D making CTR1 expression worse, where deficiency = not enough ATP7a, balance)
https://www.nature.com/articles/s41380-019-0566-2#Sec7 intake of beta-carotene was demonstrated to be low in some schizophrenia cohorts
https://pubmed.ncbi.nlm.nih.gov/24434091/
^ here a synthetic **vitamin a** drug was used with positive effect in people with schizophrenia
too much vitamin A maybe could skew things away from CTR1 more by countering vitamin D . so personally i would like a closer ratio of vit D to A than normal considering importance of ctr1 here. maybe even 1:1 idk its guess work. beef liver is good source of vitamin A + copper , if have good thyroid hormone to process it
**Thyroid hormone** also increases ATP7a , needed for ceruloplasmin and exports copper from the liver (posted by youngsinatra before https://portlandpress.com/view-large/figure/2309956/bj4430103fig3.jpeg
**& here's a great vitamin D effect in schizophrenia in humans**
The psychotropic effect of vitamin D supplementation on schizophrenia symptoms
**(interestingly only 3.8% of people tested had vit d in normal range even though it was summer)**
"Although our study population consisted of outpatients and blood samples were taken between May and July when sunlight is more intense, the rates of low vitamin D levels were found to be high"
^-- before & after vitamin D intake for 8 weeks. impressive effect size
Our finding of decreased CTR1 in the cells and fibers of the superficial layers of the entorhinal cortex lends is consistent with the importance of copper in myelin integrity and dendritic spine formation, as well as cellular function. Deficits of myelin and associated oligodendrocytes have been well replicated in schizophrenia in several brain regions
Cortical areas exhibit decreased markers of myelin basic protein (a key component of myelin), fewer oligodendrocytes, abnormal oligodendrocyte morphology, oligodendrocyte degeneration, myelin thickness and laminar abnormalities, as well as downregulation of key myelin related genes and proteins in schizophrenia
Interestingly, application of the copper chelator cuprizone results in massive, albeit reversible, demyelination and oligodendrocyte death. indicating that copper must play a crucial role in myelin and oligodendrocyte function and integrity. **If a deficit of cuprinergic signaling and copper content exists in the superficial layers of the entorhinal cortex in schizophrenia as indicated by our finding of decreased CTR1, this could impact white matter integrity in this area.**
In contrast to our findings in the superficial layers of the entorhinal cortex, we observed significantly elevated immunolabeling of CTR1 in the molecular layer of the dentate gyrus, consistent with a trending increase of CTR1 mRNA within the middle temporal area
In further support of our suggestion of compensatory upregulation of CTR1 in the molecular layer of the dentate gyrus in response to copper deficit is the observation that the dentate gyrus exhibits deficient signalling in schizophrenia
Taken together, our data suggest dysregulation of copper transport and homeostasis in the hippocampus and entorhinal cortex in schizophrenia that is region and layer specific, a potential mechanism of schizophrenia pathology that was previously unstudied. **Deficient copper within the hippocampus could have deleterious effects, including decreased energy metabolism and neuronal signaling, deficits of myelin maintenance and integrity, and an overall pathological modulation of the trisynaptic pathway crucially involved in memory and cortical function** {seen in alzheimers} Therefore, further study is needed to assess cellular copper content and transporter status of each layer and cell type of the hippocampus in schizophrenia, as well as replication of the current study. **Abnormal copper and other trace metals are becoming increasingly associated with schizophrenia and other forms of psychosis** (Bitanihirwe and Cunningham, 2009; Joe et al., 2018; Schoonover et al., 2018) and therefore could provide a new mechanism to be targeted for development of better treatments. For instance, **treatments that mimic the functions of CTR1 might enhance copper transport from the blood to the brain parenchyma could be therapeutic.**
extra info: auditory dysfunction (hearing things) brain region:
In a comprehensive narrative review Hu et al. [30], summarized studies of glutamate neuron morphology, of the synthesizing and metabolizing enzymes for glutamate and its co-agonists and of the expression of glutamate transporters and receptors. They reported clear evidence in several regions, from multiple labs of reduced dendrite length and complexity and of lower levels of synaptophysin, a marker of axon boutons.
Additionally, reduced pyramidal somal volume has been found predominantly in layer III of dorsolateral prefrontal cortex (DLPFC) and auditory cortex. Hence, the dendrites, axons and cell bodies of cortical glutamatergic neurons are clearly reduced in size in schizophrenia.
www.nature.com
Reduced Pyramidal Cell Somal Volume in Auditory Association Cortex of Subjects with Schizophrenia - Neuropsychopharmacology
Subjects with schizophrenia have decreased gray matter volume of auditory association cortex in structural imaging studies, and exhibit deficits in auditory sensory memory processes subserved by this region. In dorsal prefrontal cortex (dPFC), similar in vivo observations of reduced regional...
^ the reduced volume in auditory cortex is mainly because of smaller cell size rather than fewer cell numbers
**Volume decreases were found in** 62% of 37 studies of whole temporal lobe, and in **81% of 16 studies of the superior temporal gyrus (and in 100% with gray matter separately evaluated).** https://www.biologicalpsychiatryjournal.com/article/S0006-3223(99)00018-9/fulltext00018-9/fulltext)
so less gray matter volume of superior temporal gyrus of temporal lobe (where auditory cortex is), common to schizophrenia, and seems the volume drop is mainly due to smaller cell size instead of fewer cells

2024.06.05 10:32 muzso Loading custom (AAOS) applications on the infotainment

I've found this post at XDA: https://xdaforums.com/t/help-sideload-apps-polestar-2.4383145/
A member wrote that he could put a number of open-source apps on his Polestar 2 via the Google Play Console's internal testing feature.
This essentially means that:
  1. You register a Google developer account (requires a one-time 25$ fee).
  2. Compile an app targeting the AAOS (Android Automotive OS).
  3. Upload it to the (Google) Play Console, but don't send it in for publishing!
  4. Start an internal test where you can make the app available for a limited number of Google accounts.
  5. Test it in an Android Studio emulator.
  6. Install it on the car's infotainment (e.g. EX30) via the Google account that you use in the car.
I've tested this and it works as advertised.
I've taken an official Hello World app (from https://github.com/android/car-samples/tree/main/car\_app\_library/helloworld), compiled it with Android Studio, uploaded it to the Play Console, shared it via Internal Testing with my car's account and voila. The only single glitch was that the account used in the car needed "age verification", which you can do by visiting the https://myaccount.google.com/age-verification link (of course using the account for which you want to do it :) ).
Here's a video of the Hello World app running: https://www.youtube.com/shorts/5zJDHWykqY4
I've already prepared a list of open-source apps that I'll try to port to my EX30.
The XDA member published his own list that he already tested: https://xdaforums.com/t/help-sideload-apps-polestar-2.4383145/post-87120195
E.g. one thing that I miss from the EX30 is the capability to track the car's whereabouts. Since the infotainment system is all Google, it'd make sense the Google's location sharing can be used for this. But unfortunately it's not available in the Google Maps app in the car. :( So I'll just install the Traccar client and use my Traccar server for this purpose. I previously used Traccar (https://www.traccar.org/) to track down our dog (with a GPS tracker), but Traccar's original goal is/was to track cars. So that's what I'll use it for. :)
P.S.: this is not EX30 specific! Usually an AAOS app runs on all/most AAOS infotainment systems, so if somebody makes an app available via internal testing, it'll run on a Polestar, an EX30, an XC40, etc.
So pretty much any car with GAS (i.e. Google Play, etc.): https://en.wikipedia.org/wiki/Android_Automotive#Vehicles_with_Android_Automotive_(with_GAS))

2024.06.05 08:47 delbertgrady1921 Did I have encephalitis?

When I was in college in 2016 I got very sick. There was a mumps virus going around school and myself and a friend both fell ill. It was the worst period of my life. The first thing that happened was extreme nausea but without vomiting. I remember being so nauseous that I tried to make myself puke but couldn't. Next I started having extreme light sensitivity. When I walked outside one morning it physically hurt to keep my eyes open. I ended up going to a clinic and they did blood tests. Told me I was sick with a viral infection and my body needed to fight it off. I was not running a fever. No other tests.
The next few days I could barely sleep, and I felt extreme confusion and disorientation. I would wake up with my legs shaking uncontrollably and I'm not sure if these were seizures or what. The problem is at this point I started taking benadryl for sleep and I had never taken this before. I don't know if my body had a reaction to it but I ended up having horrible waking nightmares and tinnitus, even auditory hallucinations. I was terrified and didn't know what was happening at all. My mouth was endlessly dry, no matter how much water I was drinking it felt like my tongue would just stay dry like sandpaper.
I ended up going back to the clinic and they put me on an IV as I was very dehydrated. Again no other tests aside from more blood samples. I was having panic attacks and got severe derealization from the whole thing that lasted months. I remember feeling like I didn't recognize myself at all and the world felt surreal and terrifying for some time. I was worried I had permanent brain damage and frankly I've never felt 100% since then. I also have an anxiety disorder which heightened after this whole incident.
I've always wondered if I had untreated viral encephalitis, or if that's a ridiculous notion. As far as I know I never ran a fever, or at least nothing severe. I don't remember feeling any headaches or pressure. Just a lot of cognitive changes.

2024.06.05 08:45 Pugg34 Possibly lost song from a joke game, not sure [unidentified media]

I am not sure how to describe this but I made a post on watzatsong. Basically I was achievement grinding on a game called "Grounded Sim: Steam Edition" its a horror game about those old Cailou Gets Grounded Youtube videos from the early 2010's. While achievement hunting I noticed one that seemed really easy known as "The Fun Will Never End!" to unlock said achievement you must click on sound test and spam the crap out of the button in the middle, from there a mask will go up and next thing you'll see is a weird bunny guy wearing the mask dancing next to main character. In the background this song plays but unfortunately is slowed down extremely. It's hard to decipher the lyrics if they are in English or what but I think it goes, "Gling Gling, all that I need"
I kinda like older style of music, plus I was curious on this song. I can't find the files at all which sucks but fear not as I recorded it using OBS to get said song, I then put it in audacity and sped it up to a percent that seems reasonable however I am not sure if the song is at the right speed or tempo. I tried to use Shazam however it didn't detect it slow or at the edited speed. So now I am going to the depths of the internet and trusting you all to help on this journey, I only have a 30 second clip since I am taking from watzatsong LOL. But I will try to upload a longer version if need be but it sounds like a song from the 60's or 70's.
Anyways all, I hope you all can help me and if you all could please make sure to share throughout any other platforms. The more people hear this, the easier it is that this song that MIGHT be lost will be found. thank you all in advance for helping and please let me know if a longer clip will be better.

2024.06.05 08:17 myhandsyourbones Should I be skeptical of biopsy results?

Curious if anyone has a similar experience…
Biopsy done on 5/28 and received results from LabCorp on 5/31. Have not received follow up phone call from OBGYN yet but here’s what I received from LabCorp:
Diagnosis - LEFT LABIA BIOPSY:
SPONGIOSIS, MILD ACANTHOSIS AND CHRONIC INFLAMMATION.
PAS STAIN FAILS TO DEMONSTRATE ANY FUNGAL ORGANISMS (CONTROL STAINS APPROPRIATELY).
NO EVIDENCE OF MALIGNANCY.
—————————————————— First symptoms of possible LS appeared a year ago and 6 months ago started active treatment with Clobetabsol (daily for one month, every other day for one month, and then 2x/ week until present). Added Estradiol 2x/ week, 2 months ago.
I’m curious as to why the results of the biopsy do not explicitly state positive or negative for Lichen Sclerosus when I was under the impression that was the point of doing the biopsy.
Is this typical? If Lichen Sclerosus is not present in the biopsy sample is it just not explicitly stated in the results?
Essentially, has anyone else had a biopsy for LS and the results come back with no mention of it anywhere?! I’m just a bit worried that after all of this, it wasn’t even tested for (bah)!
Thanks! I’ve been hanging out here for about a year now and have found this subreddit/ community to be one of the most informative and gentle places on the Internet - hugs to you all!
